Materials

Caffeine (CAS #58-08-2, > 99% purity) was purchased from SigmaAldrich Co., Ltd. (St Louis, MO, USA). Safranin-O (CAS. NO. 477-73-6)and fast green (CAS. NO. 2353-45-9) were obtained from Hengyuan Biotech Co., Ltd. (Shanghai, China). RIPA lysis buffer and BCA Protein Assay kit were purchased from Beyotime Institute of Biotechnology (Haimen, China). An enzyme-linked immunosorbent assay (ELISA) kit for detecting serum corticosterone concentration was purchased from Sangon Biotech Co., Ltd. (Shanghai, China). Reverse transcription and

real-time quantitative polymerase chain reaction (RT-qPCR) kits were purchased from Takara Biotechnology Co., Ltd. (Dalian, China). The oligonucleotide primers for rat RT-qPCR genes (PAGE purification)were synthesized by Sangon Biotech Co., Ltd. (Shanghai, China).Chondroitin sulfate sodium salt from shark cartilage, DMEM/F12 (1:1)and fetal bovine serum (FBS) were supplied by Gibco (St Louis, MO,USA). Polyclonal rat antibodies including TGFβR1 (ab31013), SMAD2/3 (ab232326), SOX9 (ab185230), HDAC2 (ab213700), aggrecan (ab36861) and Col2 (ab34712) were purchased from Abcam plc.(Cambridge, Cambridge shire, UK). The histone deacetylase 2 (HDAC2)siRNA oligo was purchased from Gene Pharma Co. (Jiangsu, China).Other chemicals and agents were of analytical grade.
